Broadpharm Alkyne PEG 系列產品
Sharpless及其同事在2001年定義了click化學來描述在溫和的含水條件下快速和有選擇性地相互作用(“click”)的成對官能團。click化學概念由于其溫和的條件以及高選擇性已經擴展到生物標記,化學生物學領域和藥物研發。
總體上有三種click化學類型:
1 。銅(I)催化疊氮 - 炔click化學反應(CuAAC)
經典click化學使用銅作為催化劑,其需要適當的螯合配體,如TBTA和THPTA。TBTA涵蓋了click化學的一些實際應用,除了含水偶聯反應外,THPTA通過允許整個反應在水中運行進一步簡化click化學,從而為click反應提供生物兼容性。
2.應變促進的疊氮 - 炔烴click化學反應(SPAAC)
由于需要細胞毒性銅催化劑,CuAAC反應在應用中受到限制。一種無銅無毒的疊氮化物標記方法是應變促進疊氮 - 炔click化學(SPAAC)反應。通過使用應變環辛炔而不需要外源催化劑,SPAAC反應具有顯著降低的活化能。
3.四嗪 - 烯烴連接
Tetrazine - 烯烴連接非常適合體內細胞標記以及低濃度應用。四嗪官能化的分子可以與末端或應變的烯烴分子反應以通過二氫吡嗪部分形成穩定的綴合物,其被認為是無毒的生物分子標記方法。

1. 在磷酸鈉緩沖液中制備疊氮化物連接的抗體。
2. 通過SPAACclick化學,將疊氮化物修飾的抗體(400μg,在1mL TBS緩沖液中)與含有DBCO / DIBO / BCN部分的分子(200μL的2mM DMSO溶液)一起在室溫下優化時間。
3. 終產品可根據需要通過尺寸排阻色譜法或其他方案進行純化。
