單位定義:
TGIRT的一個單元® -III逆轉錄酶(RT)活性是酶的使用聚在60℃下聚合1納摩爾的dNTP的在1分鐘(RA)所需要的量/寡(dT)42作為底物。

酶屬性和新穎活動:
比逆轉錄逆轉錄酶更高的熱穩定性,持續合成能力和保真度,允許從高度結構化或重度修飾的RNA (例如tRNA)和包含富含GC的重復擴增的RNA合成全長,端對端cDNA 。1-9,12,15,18
新型端對端模板轉換活動,可在反轉錄過程中連接RNA-seq或PCR適配器,并且無需單獨的RNA 3'-適配器連接步驟。1這種模板轉換活性極大地促進了鏈特異性RNA-seq文庫的構建,而且比使用隨機六聚體引物或使用RNA連接酶進行銜接子連接的方法具有更小的偏差。1,7,8
從退火引物合成cDNA。將退火的引物應具有推測的T 米 > 60的直徑: C.酶與反應底物混合,在室溫下,通過加入dNTP的反應開始30分鐘的預溫育,建議。新應用的zui宜條件應該通過測試25-450mM NaCl的一系列鹽濃度來確定。

酶的優點:
1.全面的鏈特異性轉錄組分析。
核糖核碎片,通用人類參考RNA樣品的TGIRT®-seq概括了人類轉錄物和穗突起的相對豐度,與非鏈特異性TruSeq v2相比,并且優于鏈特異性Tru-Seq v3。TGIRT®-seq比TruSeq v3具有更高的鏈特異性,并消除了TruSeq固有的隨機六聚體引發的取樣偏差。TGIRT®-seq顯示出更加統一的5'至3'基因覆蓋范圍,并且比TruSeq識別更多的剪接點。TGIRT®-seq能夠在與結構化小型ncRNA相同的RNA-seq中同時分析mRNA和lncRNA,包括tRNA,TruSeq數據集基本上不存在tRNA。8

使用錨定寡核苷酸(dT)引物,可以構建RNA聚合腺苷酸化的RNA文庫,與沒有ribodepletion步驟的逆轉錄病毒RT相比,具有更均勻的5'至3'覆蓋范圍。1
通過基于毛細管電泳的方法(如SHAPE或DMS結構圖)進行RNA結構作圖,其顯著的讀數長度和更少的提前停止時間比逆轉錄病毒RTs更短。1,16

通過直接在DNA鏈的3'末端啟動DNA合成,同時連接DNA-seq接頭而不進行末端修復,拖尾或連接,以更簡單的工作流程捕獲的DNA末端。能夠分析核小體定位,轉錄因子結合位點,DNA甲基化位點和起源組織。17
